Aim

The use of cocaine is increasing worldwide and currently represents one of the most frequent indications for drug treatment. Although cocaine chronic use has been shown to lead to neurotoxicity in rodents and humans, being associated with high morbidity and mortality rates, its recreational use, which may lead to addictive behaviour, is often neglected. Materials & Methods

A total of 16 male Balb/c mice with a mean age of 6-weeks and an average weight of 26.4 g were used in this study. The animals were randomly divided into four groups according to the intraperitoneal (ip) administration of saline (0.9% NaCl), cocaine (0.5 mg/kg), quercetin (50 mg/kg) or cocaine followed by quercetin (referred dosages). 18 F-FDG was used to study cerebral metabolic rates of glucose consuption, which were reflected by local radiotracer uptake changes in brain metabolic activity...

Results

Mice exposed to cocaine showed a metabolic activity decrease mainly at Striatum (SUV St : 1.11±0.08) and Amygdala (SUV AMY : 1.74±0.13) when compared to controls (saline group, SUV St : 1.90±0.28 and SUV AMY : 3.00±0.38). In opposition, tracer uptake increase for mice exposed to quercetin (SUV St : 2.03±0.14 and SUV AMY : 4.08±0.40). Once cocaine was administered followed to quercetin, its effects were mitigated by the latter (SUV St : 2.03±0.07 and SUV AMY : 3.46±0.33). Figures 1 and 2...

Conclusion

Preclinical PET plays a fundamental role, not only in the validation of animal models for human brain disease, but also in the quantitative measurement of regional changes in brain activity. For the first time, it has been demonstrated that an acute single low dose of cocaine can induce metabolic neurological changes.
